Abbapolin 8 is a specific, non-peptidic inhibitor of PLK1 Polo-box domain (PBD), directly engages PLK1 in cells, induces selective degradation of endogenous PLK1.
Abbapolin 8 potently engages PLK1 in cells as determined by an increase in PLK1 thermal stability at sub-micromolar doses, but not for PLK3.
Abbapolin 8 inhibits PC3 prostate cancer cells with IC50 of 3.41 uM in a 72-hour MTT assay.
Abbapolin 8 inhibits colony formation of PC3 cells with IC50 of 6.28 uM.
Abbapolin 8 causes PLK1 degradation with DC50 of 5.005 uM in PC3 cells (72 h).
Abbapolin 8 induces degradation of PLK1 protein in PC3 prostate cancer xenograft tumors, shows synergy effect in combination with enzalutamide.
