hCES2A inhibitor 9d is a potent, selective and covalent human carboxylesterase 2A (hCES2A) inhibitor with IC50 of 0.12 nM and Ki of 0.43 nM.
hCES2A inhibitor 9d selectively modifies hCES2A at the catalytic serine (Ser228), thereby blocking its catalytic activity.
hCES2A inhibitor 9d shows good cell-membrane permeability and has IC50 of 19.65 nM for inhibiting intracellular hCES2A in living cells.
hCES2A inhibitor 9d (30 mg/kg and 60 mg/kg) significantly promotes ZO-1 and Claudin-1 expression in colon tissues, significantly alleviates irinotecan-induced diarrhea and dextran sulfate sodium-induced colitis.
