ZX703 is a small molecule, ML210-based glutathione peroxidase 4 (GPX4) PROTAC degrader, effectively induces GPX4 degradation with DC50 of 135 nM in HT1080 cell line, Dmax of 86%.
ZX703 exhibits HT1080 cell growth inhibition with IC50 of 435 nM.
ZX703 significantly degrades GPX4 through the ubiquitin–proteasome and the autophagy–lysosome pathways in a dose- and time-dependent manner.
ZX703 induces ROS accumulation in a dose-dependent manner, subsequently leading to ferroptosis in HT1080 cells.
