YH-0623 is a potent, selective mitochondrial RNA polymerase (POLRMT) inhibitor with IC50 of 45 nM on the 22Rv1 cell line, significantly reduces the ND1 mitochondrial factor to 14%, demonstrating 1.5-fold greater activity than IMT1B.
YH-0623 selectively reduced the expression of mitochondrial genes ND1, CytB, and COX1, while transcripts mediated by nuclear RNA polymerases I, II, and III remained unaffected.
YH-0623 induced a dose-dependent downregulation of specific subunits of respiratory chain complexes encoded by mtDNA, including NDUFB8 (complex I), UQCRC2 (complex III), and COX1 (complex IV).
YH-0623 significantly stabilized POLRMT, specifically targets mitochondrial transcription through POLRMT inhibition.
YH-0623 suppresses mitochondrial-related gene expression by inhibiting mitochondrial transcription.
