XYL-1 is a highly potent, selective PARP7 inhibitor with IC50 of 0.6 nM, shows much weaker inhibitory effect on PARP1 (IC50 >1 uM).
XYL-1 could enhance the type I interferon signaling in vitro.
XYL-1 shows higher inhibitory activity (IC50 = 0.6 nM) than that of RBN-2397 (IC50 = 6.0 nM).
XYL-1 (50 mg/kg administered orally twice daily) could significantly inhibit tumor growth (TGI: 76.5 %) in CT26 tumor-bearing mice, restores type I interferon signaling and promote T cell infiltration in tumor tissues.
