VG220 is a selective small-molecule inhibitor of p85α-cSH2-PI(3,5)P2 interaction with IC50 of 4.2 uM, promotes prolonged activation of PI3K.
VG220 does not significantly inhibit p85α-cSH2-K653A, a PI(3,5)P2 binding-compromised mutant of p85α-cSH2, does not effectively inhibit PI(3,5)P2 binding of p85α-nSH2, also not interfere with membrane binding of other membrane binding proteins, including two representative PIP3-binding PH domains from Akt1 and PDK1.
VG220 prolongs PDGF-induced PI3Kα activation by specifically blocking p85α-PI(3,5)P2 binding, thereby preventing PM displacement of PI3Kα, in WT HEK293 cells.
VG220 (5 uM) promotes prolonged phosphorylation of endogenous Akt1 at S473 (pS473) in HEK293 cells.
VG220 (10 uM) induced substantial neurite growth in mouse cortical neurons.
