UNI110 is a potent small molecule inhibitor of mismatch repair (MMR)-deficient cancer cells, selectively kills MutSa deficient cells with IC50 of 25.05 µM in HEC59 cells and 56.57 µM in HEC59-2 cells.
UNI110 treatment significantly increases γH2AX levels specifically in MutSα-deficient cancer cells and in MSH2-knockdown cells.
UNI110 impairs HR through a mechanism independent of cell cycle regulation.
UNI110 disrupts homologous recombination (HR) repair by inhibiting the MSH2-MSH3 complex, likely through disruption of the MSH2-EXO1 interaction.
