UCI-LC0019 is a specific small molecule that restores mutant p53 activity by targeting the cryptic L1/S3 pocket of p53, directly binds to mutant p53 and restores wild-type-like conformation of mutant p53R175H.
UCI-LC0019 rapidly induced apoptosis compare with APR-246.
UCI-LC0019 exhibits significantly increased solubility while maintaining activity and selectivity compare with UCI-LC0023.
UCI-LC0019 (3.3 mg/kg, 10 mg/kg, i.p.) exerted anti-tumor effect in tumor models, treatment with UCI-LC0019 resulted in a significant reduction of tumor volume in p53R175H carrying TOV112D tumors, did not significantly inhibit the growth of TOV112Dp53 / or HCT116 (p53 +/+) tumors.
