SC-3 is a synthesized tetrandrine-derived small-molecule inhibitor of two-pore channel 2 (TPC2), directly suppresses TPC2 activity enhanced by O-glycan truncation.
SC-3 robustly inhibited both basal and TPC2-A1-N–stimulated inward currents in whole-lysosomal patch-clamp recordings in glycosylation-deficient THP-1 cells.
SC-3 directly attenuates TPC2 conductance independently of the permeant ion species.
