TOP5300 (TOP05300) is a potent, selective, orally active allosteric agonist of follicle stimulating hormone receptor (FSHR) with EC50 of 9.72 nM, 80-fold selectivity over hLHR.
TOP5300 stimulated concentration-dependent increases in estradiol production in the absence or presence of a low concentration of rec-hFSH (0.2 pM) in cultured rat granulosa cells.
TOP5300 also stimulated concentration-dependent increases in testosterone production from cultured rat Leydig cells.
TOP5300 is a full agonist on the FSH receptor in human granulosa cells, stimulates estradiol production with EC50 of 474 nM.
TOP5300 (5, 10 and 25 mg/kg, p.o.) stimulated follicular maturation in immature rats.
