TED-655 is a small-molecule covalent inhibitor of TEAD-YAP1 transcription factor, reduces TEAD and YAP1 protein levels.
TED-655 binds to the TEAD lipid pocket and form a covalent bond with a conserved cysteine.
TED-655 inhibits transcriptional activity of all four TEADs.
TED-655 (10 uM) shows a subtle but concentration-dependent reduction of TEAD and YAP1 protein levels in MDA-MB-231, MPNST 90-8, and SaOS-LM7 cells.
TED-655 shows no effect on TEAD1 or YAP1 mRNA levels, suggesting that reduction in protein levels of TEAD and YAP1 is likely post-translational.
TED-655 inhibits cancer cell proliferation with IC50 in the single-digit micromolar range for NCI-H226.
