SyntOFF is a specific small molecule inhibitor of syntenin with IC50 of 37 uM in HTRF assays, targets the PDZ2 domain of syntenin.
SyntOFF does not affect Erbin‐P0071 or GRASP55‐JAM‐B complexes in HTRF.
SyntOFF displays a KD value of 130 µM for syntenin‐PDZ tandem plus C‐terminal domain.
SyntOFF decreased MCF‐7 mammosphere formation, with no effect when MCF‐7 cells is KO for syntenin expression, interferes with cell migration, proliferation
SyntOFF regulates the loading of exosomes with syntenin‐dependent cargo and EpCAM.
