Surbinsertib (STC-8123) is a potent, selective, and orally available Wee1 inhibitor with biochemical IC50 of 1.2 nM, exhibits cellular target engagement (pCDC2 MSD) in A427 cells with IC50 of 180 nM.
STC-8123 shows superior kinase selectivity compared to AZD1775 and Zn-C3 in a broad kinase panel (ScanMAX; >450 kinases).
STC-8123 exhibits anti-proliferative activity in a variety of tumor cell lines with IC50 of 290 nM, 440 nM, 300 nM, and300 nM for A427 (Lung), OVCAR3 (Ovarian), HCC1806 (Breast) and SW620 (Colon).
STC-8123 demonstrates robust anti-tumor activity and sustained target engagement in vivo in tumor models.
