Skp2 inhibitor 10h is a potent S-phase kinase-associated protein 2 (Skp2) inhibitor with IC50 of 0.38 uM against Skp2-Cks1 binding.
Compound 10h has an IC50 values of 11.24 μM and 10.32 μM against lung cancer NCl–H1299 and esophageal cancer KYSE-510 cells, respectively.
Compound 10h induces cellular accumulation of Skp2 substrates.
Compound 10h could increase the sensitivity to cisplatin in xenograft model bearing NCl–H1299 cells.
Compound 10h inhibits tumor stemness, significantly induces G2/M-phase cell cycle arrest and suppresses the protein levels of tumor stemness markers including CD44, Nanog, OCT4 and SOX2.
