SU0448 is a specific, cell-permeable small molecule activator of the DNA repair enzyme Human MutT Homolog 1 (MTH1) with AC50 value of 5.5 uM, inhibits cell viability of HCC1806 cells with IC50 of 36.1 uM.

SU0448 showed intracellular interaction with MTH1 and over 200% activation in HCC1806 cells at 20 uM, does not change MTH1 protein expression.

SU0448 reduces genomic DNA damage both in cancerous and noncancerous cells regardless of endogenous MTH1 protein level.

SU0448 has little residual off-target kinase activity, very low toxicity, and favorable permeability properties.

SU0448 can lower levels of 8-oxo-dG in cellular DNA substantially.
