Y-320 is a specific, allosteric small-molecule STING inhibitor, inhibits IL-17 production by CD4 T cells stimulated with IL-15 with IC50 of 20-60 nM, blocks STING Golgi translocation and downstream signaling.
Y-320 allosterically targets the non-canonical transmembrane domain (TMD) pocket without competing with the endogenous agonist 2′3′-cGAMP by engaging residues Y46 and W119 in the transmembrane helices 2 and 4.
Y-320 enhances TP53, DMD, and COL17A1 PTC readthrough by G418 and increases cellular protein levels and protein synthesis.
Y-320 dose-dependently ameliorates cisplatin-induced acute kidney injury in mice.
