SPRi3 (SPR inhibitor 3) is a small molecule inhibitor of sepiapterin reductase (SPR) with IC50 of 74 nM in cell-free assays, efficiently reduces biopterin levels with IC50 of 5.2 uM.

SPRi3 also significantly reduces SPR activity in mouse primary cultures of sensory neurons (IC50=0.45 uM) without affecting the activity of GCH1。

SPRi3 reduces pain hypersensitivity effectively with a concomitant decrease in BH4 levels in target tissues, acting both on sensory neurons and macrophages, with no development of tolerance or adverse effects.
