SPH7050 is a gut-restricted Lanthionine synthetase C-like protein 2 (LANCL2) agonist, binds LANCL2 with SPR KD of 2.73 uM, inhibits prostaglandin E2 (PGE2) production in bone marrow-derived macrophages (BMDMs) with IC50 of 0.85 uM.
SPH7050 binds more deeply than Omilancor (BT-11), making a hydrogen bond to GLU-213 and cation–π interactions with LYS-164 and ARG-118.
SPH7050 (50 mg/kg, p.o.) significantly alleviated TNBS-induced colitis in vivo.
SPH7050 has intestinal-restricted absorption with minimal systemic exposure.
