SCoV2 Plpro-IN-19 is a potent and selective inhibitor targeting the papain-like protease of SARS-CoV-2 (SCoV2 PLpro) with IC50 of 0.44 uM, binding Kd of 2.6 uM.
SCoV2 Plpro-IN-19 displays no inhibitory effect at 10 uM against 10 DUBs or DUB-like proteases, including USP14 and OTUB1.
SCoV2 Plpro-IN-19 significantly inhibited SCoV2 PLpro in 293T cells at 10 uM.
SCoV2 Plpro-IN-19 efficiently suppressed viral replication of with a submicromolar IC50 0.18 uM without cytotoxicity (CC50>50 uM).
