Ripasudil (K-115) is a potent, specific Rho-associated coiled-coil containing protein kinase (ROCK) inhibitor with IC50 of 51 nM and 19 nM for ROCK1 and ROCK2 respectively.
Ripasudil (K-115) significantly reduced intraocular pressure (IOP) in a dose-dependent manner in rabbits.
Ripasudil (K-115) induced retraction and rounding of cell bodies as well as disruption of actin bundles on monkey trabecular meshwork (TM) cells.
Ripasudil (K-115) significantly decreased transendothelial electrical resistance (TEER) and increased the transendothelial flux of FITC-dextran in SCE-cell monolayer permeability studies.
Ripasudil (K-115) disrupted cellular localization of ZO-1 expression in SCE-cell monolayers.
