RMC-6272 (RMC6272) is a potent, selective bi-steric inhibitor of mTORC1 with cellular IC50 of 0.44 nM for p4EBP1 inhibition, 27-fold selectivity over mTORC2.
RMC-6272 displays >1000-fold selectivity over other lipid kinases.
RMC-6272 induces widespread apoptosis and deep tumor regressions in a xenograft model of KRASG12C mutant NSCLC bearing a loss of function mutation in STK11, a negative regulator of mTORC1.
