RBPMS inhibitor RB98 is a specific small-molecule inhibitor of RNA binding protein with multiple splicing (RBPMS) with SPR KD of 402 nM, exhibits competitive inhibition of FOXO1 mRNA-RBPMS binding with IC50 of 3.1 uM in fluorescence polarization (FP) assays.
RB98 treatment of leukemia cells resulted in down-regulation of FOXO1 expression, alongside an enrichment of FOXO1 transcripts associated with RBPMS.
RB98 inhibited the proliferation and promoted the apoptosis of leukemia cells.
RB98 treatment resulted in a decrease in the FOXO1 mRNA abundance in polysomes, induced apoptosis in cells and diminished the colony-forming ability of primary AML cells.
RB98 reduced the leukemic burden in RUNX1-ETO9a–driven AML models.
