Pin1 inhibitor PiB is a specific small molecule inhibitor of parvulin family peptidyl prolyl isomerase (PPIase) Pin1 with IC50 of 1.5 uM, inhibits Par14 as potently as Pin1 with IC50 of 1.0 uM.
PiB shows limited activity against Cyclophilin C PPIase activity (IC50>10 uM).
PiB inhibited proliferation of wild-type MEF (IC50=1 uM), does not inhibit the growth of Pin1−/− MEFs.
exhibits anticell proliferation activities against cancer lines HSC2, HSC4, HCT116, Huh7, OVK2, SKOV3, and HepG2 with IC50 of 0.7-4.8 uM.
PiB (1 uM) for 48 hr showed decreased expression of cyclins D1 and B1 in HCT116 cells, increased the population of cells in S phase at 48 hr after cells.
Pin1 inhibitor PiB protects rats against lipopolysaccharide-induced respiratory inflammation.
