PTX-BD4-3 is a specific small-molecule, non-peptide TrkB partial agonist, supports the survival of cultured hippocampal neurons with EC50 of 300-500 nM.
PTX-BD4-3 promotes survival of 3T3-TrkB cells in a dose-dependent manner and to a similar degree to the parent compound, LM22A-4, has no effect on survival of TrkA-, TrkC- or p75-expressing cells.
PTX-BD4-3 promotes the recruitment of the TrkB adaptor protein Shc (also known as Shc1) to TrkB, triggers activation of TrkB.
PTX-BD4-3 (50 mg/kg, i.p.) significantly increased pTrkB/TrkB and pAKT/AKT in null male Mecp2 mutant mice, and had no significant effect on pERK/ERK.
PTX-BD4-3 (5 mg/kg) reduces apneic breathing in heterozygous (Het) female Mecp2 mutants.
PTX-BD4-3 restores motor learning in Het female Mecp2 mutants.
