PF-07265028 is a potent, selective hematopoietic progenitor kinase 1 (HPK1/MAP4K1) with Ki of <0.05 nM, and cellular IC50 of 17 nM (pSLP76 inhibition).
PF-07265028 only inhibits at a less than 30-fold window relative to HPK1 for 2 out of 395 kinases (MAP4K5 and TAOK1).
PF-07265028 shows potent functional response in human T cells as measured by induction of IL-2 production with EC50 of 59 nM.
