P053 (P-053, CerS1 inhibitor P053) is a potent, selective, orally active inhibitor of ceramide synthase 1 (CerS1) with IC50 of 0.5 uM, displays >10-fold selectivity for inhibition of CerS2, CerS4, CerS5, and CerS6.
P053 (P-053, CerS1 inhibitor P053) selectively reduces C18 sphingolipids in cultured cells, with no effect on HEK293 cell viability at or below 1 uM.
P053 (P-053, CerS1 inhibitor P053) increases fatty acid oxidation in skeletal muscle and impedes increases in muscle triglycerides and adiposity in mice fed a HFD, but does not protect against HFD-induced insulin resistance.
P053 (P-053, CerS1 inhibitor P053) increases palmitate oxidation and mitochondrial respiratory capacity in SkM.
