Odemsertib (R835) is a potent, selective dual inhibitor of IRAK1/IRAK4 with IC50 of 15/14 nM respectively.

R835 selectively inhibits IRAK4 auto-phosphorylation, IRAK1 protein degradation, and IRAK4/1-specific downstream activation of the NFκB and MAPK pathways in cells.

R835 inhibits TLR4 dependent IRAK1 protein degradation in THP-1 stimulated with LPS.

R835 inhibits phospho(p)-IKK, p-NFkB, p-p38 and p-JNK with estimated IC50 of 16 nM.

R835 completely and dose-dependently inhibited TLR4-induced IL-23 production by human THP1 cells with IC50 of 81 nM, IL-1R-induced IL-6 production by HUVEC with IC50 of 51 nM.

R835 potently blocked TLR4/nigericin-dependent NLRP3 inflammasome priming, activation, and pyroptosis in human monocytic cells.
