NSH76 is a potent and selective RNA polymerase I (Pol I) inhibitor, inhibits Pol I-specific transcription initiation factor RRN3, selectively inhibits Pol I transcription by disrupting the Pol I pre-initiation complex.
NSH76 specifically binds to RRN3.
NSH76 selectively inhibits Pol I transcription in cancer cells, effectively suppresses rRNA synthesis, disrupts PIC formation, and induces nucleolar stress, in RRN3-high cancer cells.
NSH76 does not affect cMyc expression, a Pol II-driven transcript, confirming its specificity.
NSH76 induces anti-proliferative effects via p53 activation, cell cycle arrest, and apoptosis, with IC50 of 7.52, 3.73, and 4.04 µM in A549, PC3, and MCF7 cell lines.
NSH76 enhances cisplatin and doxorubicin sensitivity in resistant cells.
