NJNU-0069 is a specific small-molecule inducer of major histocompatibility complex class II (MHC-II), directly binds to the MAT domain of FASN (Kd=32.1 uM).
NJNU-0069 potently elevated the expression of MHC II genes and upregulated the surface MHC-II protein levels in a dose-dependent manner in cancer cells, including 4T1, MCF-7, and Hs578T.
NJNU-0069 is an analog of pristane, exhibits about 26-fold higher binding affinity to MAT than pristane did (NJNU-0069 Kd: 32.1 μM vs. pristane Kd: 840.6 μM).
NJNU-0069 (100 mg/kg for four times every other day) can effectively inhibit the growth on Balb/c mice bearing 4T1 tumors, fires up the tumor microenvironment by promoting immune detection.
