Lvguidingan is a piperine derivative and antiepileptic candidate with broad-spectrum anticonvulsant activity, inhibits Nav1.2 with low-micromolar potency (IC50=6.16 uM) and exhibits differential inhibitory effects across voltage-gated sodium channel subtypes.
Lvguidingan inhibited Nav1.2 in a state-dependent and use-dependent manner and altered channel gating.
Lvguidingan suppressed hippocampal neuronal excitability. inhibited epilepsy-associated SCN2A gain-of-function variants.
