Lepzacitinib (ATI-1777) is a potent, selective and ATP-competitive JAK1/3 inhibitor with IC50 of 1.5/3.6/7.1/19.0 nM for JAK1/JAK2/JAK3/TYK2, respectively.
ATI-1777 displays 32 to 833 fold selectivity for Jak1/3 over a panel of 214 human protein kinases.
ATI-1777 demonstrates inhibition of STAT phosphorylation in isolated PBMCs with IC50 of 19 nM, 5-fold selectivity over Jak1/2 and approximately 42- to 47-fold selectivity over Jak2.
ATI-1777 application inhibits IL-15–induced CCL8 expression in pig skin by 83%, selectively inhibits Jak1/3 with limited systemic exposure and without any adverse effects.
