LN5P45 is a selective, covalent, irreversible inhibitor of ubiquitin thioesterase OTUB2 with IC50 of 2.3 uM, strongly induces monoubiquitination of OTUB2 on Lys31.
LN5P45 is about 20-fold more potent toward OTUB2 over OTUD3 (IC50=56 uM).
LN5P45 also shows no activity against the closest family members OTUB1, OTUD1, OTUD2, OTUD3, OTUD6B, and Cezanne, and DUBs UCHL1 and USP16 (IC50>100 uM).
LN5P45 (10 uM) displays clear inhibition of GFP-OTUB2 in HEK293T cells, LN5P45 engages endogenous OTUB2 and is selective for OTUB2 over other ubiquitin machinery components.
