IRBM-Z-1 is a non-competitive allosteric inhibitor of ZIKV NS3 protease with IC50 of 1.8 uM (wt NS2B-NS3 protease), exhibits ZIKV antiinfectivity activity in cell culture assays with EC50 of 6.25 uM.
The I156T mutation in NS3, strongly reduced the inhibitory effect of IRBM-Z-1, in both the enzymatic and replicon assays.
IRBM-Z-1 exhibited a dissociation constant (KD) of 1.52 uM for ZIKV NS3 protease.
