IMB-9C is a specific small molecule inhibitor of heptad repeat 1/heptad repeat 2 (HR1/HR2) interaction, inhibits SARS-CoV-2-S-mediated cell-cell fusion.
IMB-9C exhibits a dose-dependent inhibitory effect on viral entry with EC50 of 32.15 uM.
IMB-9C demonstratesenhanced potency against Omicron variants compared with WT and previous variants.
IMB-9C inhibits virus-induced CPE for WT, Beta, Delta, Ba.1, Ba.2, Ba.4 and Ba.5, with IC50 values of 0.4 -3.1 uM, with low toxicity on mammalian cells such as HEK-293T, Huh-7 and Vero cells.
IMB-9C is able to block 6-HB formation between HR1 and HR2, binds with both HR1 and HR2 with SPR KD of 0.567 uM and 36.8 uM respectively.
