IKZF2 degrader 55 is a potent, selective IKZF2 degrader with DC50 of 1.8 nM by FACS assay in the Jurkat cells, Dmax=92%, shows minimal off-target activity against IMiD neo-substrates.
IKZF2 degrader 55 does not degrade a number of other neo-substrates, such as IKZF3, SALL4, and GSPT1.
IKZF2 degrader 55 displays moderate degradation against SALL4 (DC50 = 9 nM, Dmax = 57%).
IKZF2 degrader 55 at 10 and 30 mg/kg depleted IKZF2 protein by 88 and 79%, respectively, in spleen tissue, and 79 and 83%, respectively, in thymus tissue at the 6 h time-point.
