    IHCH-8110 is a potent, selective, non-brain-penetrant, peripheral-restricted serotonin 2A receptor (5-HT2AR) agonist with Ki of 47.86 nM, inhibits CRC progression by activating 5-HT2AR on enteric glial cells.
    IHCH-8110 acts as an agonist at both 5-HT2AR and 5-HT2CR in functional assays, including calcium flux and β-arrestin2 recruitment, also functions as a 5-HT2BR antagonist.
    IHCH-8110 exhibits no detectable functional activity at other GPCR family 5-HT receptors or dopamine receptors.
    IHCH-8110 suppresses CRC progression by enhancing CD8+ T cell antitumor functions.
    IHCH-8110 targets enteric glial cells (EGCs) expressed on EGCs to inhibit tumor growth.
    IHCH-8110 enhances the recruitment and polarization of CD8+ T cells by upregulating EGC-derived CXCL10 and IL-18.
    IHCH-8110 sensitizes immune-cold CRC to anti-PD-1 therapy.
