GLA-1-1 is a small-molecule agonist of large-conductance Ca²⁺-activated K⁺ (BK) channels with EC50 of 3.4 uM, induces autophagy flux by promoting autophagosome-lysosome fusion.
GLA-1-1 reduces autofluorescent deposits and RPE abnormalities in Lamp2a-/- mice as detected by FAF, OCT, and confocal microscopy.
GLA-1-1 preserves retinal structure, RPE morphology, and visual function in Lamp2a-/- mice.
GLA-1-1 attenuates Lamp2a deficiency-associated lipid accumulation in the RPE and reduces Bruch's membrane thickening.
GLA-1-1 restores lysosome-dependent degradative abnormalities in CMA-deficient Lamp2a-/- mice.
