Ferristatin II (NSC 8679, NSC8679, Chlorazol Black) is a small molecule iron transport inhibitor that promotes degradation of transferrin receptor-1 in vitro and in vivo, through a nystatin-sensitive lipid raft pathway.
Ferristatin II reduces intestinal (59)Fe uptake, lowers serum iron and transferrin saturation, with no effect on liver non-heme iron stores.
Ferristatin II decreases expression of Smad downstream targets Smad7 and Id1 and increases expression of Stat3 downstream targets, induces hepatic hepcidin expression in vivo and in vitro.
