Feeblin (IRF5 inhibitor C5) is a small molecule inhibitor of the proinflammatory TLR7/8/9-IRF5 pathway (IC50=1.6 uM) by disrupting the SLC15A4-TASL adapter module, Feeblin is an IRF5 pathway-specific inhibitor.
Feeblin (IRF5 inhibitor C5) displays specific inhibition of IRF5, shows no or little activity on STING, NOD1/2 or RIG-like helicase pathways.
Feeblin (IRF5 inhibitor C5) significantly reduces IRF5 nuclear translocation, without affecting the activation of NF-κB p65.
Feeblin (IRF5 inhibitor C5) locks SLC15A4 in a TASL binding-incompetent lysosomal outward-open conformation.
