FZ581 is a potent, pan-species inhibitor of soluble epoxide hydrolase (sEH) N-terminal phosphatase domain (sEH-P, sEH NTD) with IC50 of 59 and 490 nM for human and mouse sEH NTD, shows little to no activity against full-length sEH protein (sEH FL).
FZ581 shows high selectivity over the soluble epoxide hydrolase activity (sEH-H).
FZ581 does not exhibit significant binding toward other 9 tested phosphatases, as well as COX-1 and COX-2, lipid phosphate phosphatases (LPPs) LPP1-3.
FZ581 exhibits inhibitory activity across different species in vitro.
FZ581 shows dose-dependent inhibition of 1-oleyl monoacyl glycerol (sn1-18:1 MAG) formation through inhibition of recombinant mouse sEH.
FZ581 (1 mg/kg) demonstrated a significant increase of main sEH-P substrate levels without affecting plasma levels of sEH-H substrates in mice and rats.
