FM029 is a direct FOXP3 targeting small molecule that binds to FOXP3 (SPR KD=1.13 uM), stabilizes the FOXP3-T-bet interaction to represses T-bet-driven IFN-γ production.
FM029 binds mouse FOXP3WT protein and mouse FOXP3V408M protein, with KD values of 0.96 μM and 0.94 μM, respectively.
FM029 significantly reduced IFN-γ production in V408M iTregs but not in WT iTregs, does not suppress IFN-γ production in FOXP3-deficient Tconv cells.
FM029 markedly enhanced the ability of both WT and V408M Tregs to suppress IFN-γ production by Tconv cells.
FM029 alleviates IFN-γ+ Treg-driven tissue inflammation in V408M mice.
FM029 restrains Treg-derived IFN-γ production and ameliorates colitis.
