FAK PROTAC D4 is a potent and highly selective FAK PROTAC degrader with DC50 of 1.08 nM in PA-1 cells.
D4-mediated degradation of FAK depends on ligand binding and ubiquitin–proteasome system.
D4 inhibits cell proliferation, colony formation, cell migration, and invasion in vitro.
D4 promotes MHC-I expression and antigen presentation on the surface of tumor cells.
D4 facilitates the activation of antigen-specific CD8+ T cells.
D4 suppresses H22 tumor progression and facilitates CD8+ T cell-mediated immune surveillance.
