Lotixparib hydrochloride (DM5167 hydrochloride) is a potent, selective PARP1 inhibitor with IC50 of 0.41 nM, displays >300-fold preference for PARP1 over PARP2 in in vitro enzymatic and DNA capture assays.

DM5167 shows higher selectivity than AZD5305 in the enzymatic assay.

DM5167 efficiently reduces growth of triple-negative breast cancers.

DM5167 achieved a 45.8% tumor growth inhibition at a dose of 50 mg/kg in orthotopic xenograft model using triple-negative breast cancer cell lines.
