DKFZ-748 (DKFZ748) is a potent, specific HDAC10 inhibitor with cellular IC50 of 22 nM, >500-fold selectivity over other HDAC isozymes.
Inhibition of HDAC10 activity via DKFZ-748 (5 uM) suppressed colon cancer cell growth rescue by N8-AcSpd in HCT116 WT cells.
DKFZ-748 treatment effectively prevented restoration of polyamine homeostasis by N8-AcSpd in HCT116 cells.
DKFZ-748 showed dose-dependent growth inhibition of HeLa cells in a polyamine-limiting in vitro tumor model.
DKFZ-748 is a valuable chemical probe for studies on the enigmatic biology of HDAC10 and acetylated polyamines in both physiological and pathological settings.
