DDO-7432 is a specific, orally bioavailable molecular glue that induces aberrant STING oligomerization, inhibits STING activity with IC50 of 1.37 uM in IRF luciferase reporter assays in THP-1-Dual cells, induces self-association of STING to inhibit STING-dependent signal transduction.
DDO-7432 suppresses the inflammatory response induced by HSV-1 infection in THP-1 cells.
DDO-7432 (60 mg/kg administered intragastrically) partially suppressed IFN-β secretion in serum and downregulated the expression of Mx1 and Cxcl10 in C57BL/6JSTING-Hu mice.
DDO-7432 (30 or 60 mg/kg, oral gavage) significantly reduced ISGs expression and attenuated inflammatory parameters in muscle tissue from C57BL/6JSTING-Hu myositis model.
