CW15337 is a potent and highly selective inhibitor of NF-κB-inducing kinase (NIK) with Ki of 25 nM in biochemical assays, does not inhibit IKKα or IKKβ kinase activity.
CW15337 downregulates Bcl-XL and increases sensitivity to venetoclax against esistant CLL cells.
The combination of CW15337 and ABT-199 (venetoclax) reversed the drug resistance observed when treating tumor cells with ABT-199 alone.
CLL cells derived from patients showed increased nuclear p52 and high sensitivity to CW15337.
