CW-3308 is a potent, selective, and orally bioavailable BRD9 PROTAC degrader, effectively reduces the BRD9 protein by 68% at 1 nM (DC50 < 1 nM) and 90% at 10 nM in the G401 rhabdoid tumor cell line.
CW-3308 also effectively induced depletion of BRD9 protein in the HS-SY-II synovial sarcoma cell line in Western blotting analysis with DC50 < 10 nM and Dmax > 95%.
CW-3308 effectively inhibited cell growth in the G401 rhabdoid tumor cell line and the HS-SY-II synovial sarcoma cell line and is much more potent than BI-7273 in both cell lines.
CW-3308 50 mg/kg effectively depleted BRD9 in the tumor tissue by >90% at 3 h time point and by 45% at 24 h time point in the HS-SY-II synovial sarcoma xenograft tumor model.
