CVN636 is a potent, selective, CNS penetrant and allosteric agonist of mGluR7 with EC50 of 7 and 2 nM for human and mouse mGluR7, respectively.
CVN636 displays no clear activity agaonist other mGluRs (mGluR2,6,8 cAMP, mGluR1,3,5 Gαqi5 calcium flux assays) at 10 uM.
CVN636 (0.3, 1, 3 mg/kg oral) reduced ethanol self-administration in msP rats of alcohol use disorder.
