CF3CN is a brain permeable, synthetic 7,8-DHF derivative and TrkB agonist, robustly binds to the LCM/CC2 motif on TrkB extracellular domain (ECD) with Kd of 80.2 nM, activates TrkB and its downstream signaling pathway.
CF3CN displays a dose-dependent protection against staurosporine-induced apoptosis in T48 cells with EC50 around 26.4 nM.
CF3CN protects BR6 cells from STS-induced cell death with IC50 of 103 nM, but not TrkB-deficient SH-SY5Y cells.
CF3CN activates p-TrkB and promotes cell survival in a TrkB-dependent manner.
CF3CN dose-dependently decreases Aβ42, prevents the synaptic loss in the hippocampus in 5xFAD mouse brains.
CF3CN alleviates inflammation, inhibits delta-secretase (AEP, δ-secretase) activation, and reduces the concentrations of AEP-derived APP fragments and Tau fragments.
