Balinatunfib (SAR441566) is a potent, oral inhibitor of tumor necrosis factor receptor 1 (TNFR1) signaling, binds to human TNF with SPR Kd of 15.1 nM.
SAR441566 potently inhibits zymosan-activated CD11b expression on granulocytes in human whole blood.
SAR441566 inhibited the expression of CD11b in zymosan-stimulated human whole blood with an IC50 of 35 nM.
SAR441566 occupied human TNF with an OCC50 for total compound (includes protein bound) of 14.3 nM.
Balinatunfib (SAR441566) stabilizes an asymmetrical form of the soluble TNF trimer, compromises downstream signaling and inhibits the functions of TNF in vitro and in vivo.
